Overview | ChIP-chip (ChIP-on-chip) is a high content approach that combines chromatin immunoprecipitation (ChIP) with microarrays (chip) to analyze how regulatory and structural proteins, such as transcription factors and histone subunits, bind and interact with the genome of living cells. Through the immunoprecipitation of these protein-DNA complexes, associated sequences are pulled out of solution for analysis on DNA microarrays. Using high quality antibodies, post-translational modifications to these sequences can also be detected. The combination of ChIP and promoter or genomic tiling microarrays allows genome-wide identification of DNA-binding sites for chromatin-associated proteins with precise resolution at a variety of locations. This more comprehensive approach allows for greater insight into gene regulatory networks and can facilitate the creation of genome-wide maps of protein-DNA interactions or histone modifications.
