Data Examples
Western Blot | Detection of Human STAT6 by Western Blot. Western blot shows lysates of U937 human histiocytic lymphoma cell line untreated (-) or treated (+) with 100 ng/mL Recombinant Human IL‑4 (Catalog # ) for 30 minutes. Gels were loaded with 20 μg of cytoplasmic (Cyto) and 10 μg of nuclear extracts (Nuc). PVDF membrane was probed with 0.2 µg/mL Rabbit Anti-Human/Mouse STAT6 aa 342‑640 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF21671) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # ). A specific band for STAT6 was detected at approximately 110 kDa (as indicated). For additional reference, duplicate lysates were probed with 1.0 µg/mL Human Phospho-STAT6 (Y641) Antigen Affinity-purified Polyclonal Antibody (lower panel, Catalog # ). Background: STAT6
Signal Transducer and Activator of Transcription 6 (STAT6) is a member of the Jak/STAT signal transduction pathway and mediates cytokine signaling by IL‑4. IL‑4 activation leads to tyrosine phosphorylation and accumulation in the nucleus of STAT6 homo and heterodimers. STAT6 binds to the consensus DNA binding site TTCN4GAA. STAT6 transcriptional activation occurs by binding p100 and p160 class (NcoA1, Src-1) co-activators. Gene expression studies indicate that STAT6 mediates cell proliferation and anti-apoptotic effects of IL‑4. Also, knockout studies in mice suggest that STAT6 functions in T helper 2 (Th2) cell differentiation and transcriptional expression of cell surface markers and class switch of immunoglobulins.
