Recombinant Human His6-Ufc1 is a member of the UFM1-conjugating (E2) enzyme family that receives UFM1 from a UFM1-activating (E1) enzyme and subsequently interacts with a UFM1 ligase (E3) to conjugate UFM1 to substrate proteins. Reaction conditions will need to be optimized for each specific application. We recommend an initial Recombinant Human His6-Ufc1 concentration of 0.1-1 μM.

Ubiquitin-fold Modifier Conjugating Enzyme 1 (Ufc1) has been identified as an E2-like conjugating enzyme for Ubiquitin Fold Modifier 1 (Ufm1) (1). Ufc1 is a 116 amino acid (aa) protein with a predicted molecular weight of 19 kDa. Human Ufc1 exhibits 97% and 98% aa identity to its mouse and rat orthologs, respectively. The process of attaching Ufm1 to target proteins is termed ufmylation. It is thought to occur in a manner analogous to the ubiquitin system, including stepwise enzymatic reactions involving Ubiquitin-activating (E1)-, Ubiquiting-conjugating (E2)-, and Ubiquitin Ligase (E3)-like enzymes. In addition to Ufc1, other members of the Ufm1-conjugation system include the E1-like enzyme Uba5, E3-like enzyme Ufl1, and deconjugating enzymes UfSP1 and UfSP2 (1-3). This protein contains an N-terminal His6 tag.
